Research Interest

Maura Boldrini, MD, PhD is an Associate Professor of Psychiatry, Director of the Quantitative Brain Biology (BrainQUANT) Institute, and the Human Neurobiology laboratory in the Department of Psychiatry at the Vagelos College of Physicians and Surgeons, Columbia University Irving Medical Center, and the New York State Psychiatric Institute. Her research focuses on understanding underlying mechanisms of behavioral, cognitive, and emotional changes associated with brain diseases.

The BrainQUANT Institute’s mission is to apply cutting edge quantitative neuroscience methods to human brain tissue to understand how the brain functions and diseases develop. Quantitative cutting-edge technologies are applied to interrogate, at a single cell and regional level, the molecular basis of brain biology and pathology.

She currently has three R01s, which only 4% of all investigators achieve. Her lab focuses on single cell and spatial multiomics methods to understand genetic and epigenetic factors that shape brain cell function in psychiatric disorders, suicidal behavior, aging, dementias and COVID brain. She fostered collaborations with the bioengineering departments of Columbia and Yale. Together they applied new spatial multiomics technologies to intact human brain tissue, which produced two Nature papers: Spatial epigenome–transcriptome co-profiling of mammalian tissues and Spatial profiling of chromatin accessibility in mouse and human tissues. She is working with induced pluripotent stem cells (iPSC) and organoids to generate hippocampus neurons and test molecular effects on their differentiation, electrical activity, and survival.  Combining these technologies, she aims to identify new molecular treatment targets to improve the life of those suffering from brain diseases.

Since she became faculty in 2008, Maura’s lab has been continuously funded by four different NIH institutes (NIMH, NIA, NINDS, NIAID) and private foundations. Besides being a mentor in the Neurobiology and Behavior program and T32 Research Fellowships, she has been helping young faculty with their new R01 applications offering her research insights and pilot data, and she has been an advisor on K awards.

I am currently involved as Principal Investigator or Co-Investigator in research projects studying:

1. The relationship between trophic factors expression in the hippocampal dentate gyrus (DG) and its correlation with the amount of neurogenesis and angiogenesis in the human brain.

2. Human adult neurogenesis in normal subjects and individuals with mood disorders and the effects of antidepressant treatment on human adult neurogenesis.

3. Molecules controlling cell proliferation, maturation and survival in depressed suicide compared with non-suicide depressed individuals and controls without psychiatric diagnosis.

4. Angiogenesis and cell proliferation in the postmortem temporal cortex of autism donors and matched normal developing individuals.

Maura is one of the foremost experts in the World on adult human neurogenesis. She has been pioneering researcher on new neurons in human brain, showing persistent neurogenesis into the eighth decade of life in neurotypical subjects, blunted neurogenesis in major depression, and improved levels with antidepressant treatment. Her research on the neurobiology of mood disorders and suicide, includes investigating the effects of inflammation on neurodegeneration, and the consequences of COVID-19 infection. She published on the role of the raphe nucleus serotonergic system in sudden death in epilepsy, and the role of inflammation in major depression and COVID-19 neuronal damage. Her latest and current R01s (two from NIA and one from NIMH) focus on developing and applying new cutting edge spatial and single cell multiomic technologies to decipher the neurobiology of major depression and suicide, neurotypical aging, and Alzheimer’s Disease.

Maura’s primary collaborations within Columbia University are with Dr. Lewis Brown, Director, Quantitative Proteomics Center in the Biological Sciences Department, Dr. Erin Bush at the Columbia Genome Center, Dr. Kam Leong in the Bioengineering Department, Dr. Henry Colecraft in the Department of Physiology and cellular Biophysics, Drs. Peter Canoll, James Goldman and Andrew Teich in the Department of Pathology and Cell Biology, Dr. René Hen, Departments of Psychiatry, Neuroscience and Pharmacology, Dr. Barbara Corneo, Director, Columbia Stem Cell Core Facility, Dr. Andrew J. Dwork, Departments of Psychiatry, and Pathology and Cell Biology, Drs. Gorazd B. Rosoklija, Mark D. Underwood and myself in the Department of Psychiatry, Drs. Hanga Galfalvy, Wenpin Hou and Ying Liu in the Department of Biostatistics, and Dr. Raju Tomer in the Biological Sciences Department.

Her external collaborations are with Dr. Rong Fan, at the Yale School of Engineering and Applied Sciences, Dr. Amar Sahay at Harvard University, Dr. Robyn Klein at Washington University in St Louis, Dr. Michael Bonaguidi at the University of South California, Dr. Maria Oquendo, at University of Pennsylvania, Dr. Enrique Baca Garcia, Department of Psychiatry and Autonomous University of Madrid, Spain, and Dr. Sandrine Tourette at the University College of London.
